The native helical symmetry of uncondensed f-actin filaments is not necessarily the preferred symmetry of bundled filaments, which will depend on the particular bundling mechanism and the kind of condensing agent used. In order to explore the roles of filament twist and electrostatics in the process of bundle formation, we have carried out a series of SAXS measurements at Sector 34 ID-C on systems of bundled f-actin, using several kinds of condensing agents, such as multivalent ions, globular cationic proteins, and specific actin linking proteins. Further SAXS studies will be carried out on F-actin bundles stabilized by a variety of F-actin linking agents. We have also developed micro-fluidic devices used for aligning samples of condensed biomolecules.
